Thermostable β-Galactosidase from Bacillus acidocaldarius and Its Immobilization

Bacillus acidocaldarius HM-1 and 11-1O were selected as sources of thermostable β-galactosidase from a survey of thermophilic bacteria. Both enzymes were found to be inducible. The optimum temperature was 65℃ and 70℃ for the enzyme from strains HM-1 and 11-1O, respectively. The enzyme from strain 11-1O was immobilized on Dowex MWA-1 and Sepharose 4B and within polyacrylamide gel. The pH profiled of the enzyme imobilized on Dowex MWA-1 shifted towards the acidic side. The optimum temperature of the enxyme immobilized on Sepharose 4B became 5℃ higher compared with that of native enzyme. The half-life of the enzyme immobilized on Sepharose 4B in continuous column operation was 42 days at 50℃ when lactose solution at pH 6.5 was passed through the column and it was 32 days when bovine milk was passed. Bothe values were higher than those of the immobilized enzyme from Aspergillus niger (Lactase LP).
