Effect of Mass Transfer on Operational Stability of Immobilized Enzyme

Investase, β-galactosidase, tryopsin, and hexokinase were immobilized in polyacrylamide gel. Operational stabilities of these immobilized enzymes were measured in gels of different particulate diameter. The larger the gel diameter was, the longer the half-life of the immobilized enzyme was. This was interpreted in terms of intraparticle diffusion. From the results with invertase covalently bound to Sepharose 4B, interparticle diffusion was also found to influnece operational stability. A new equation was proposed to describe the degree of immobilized enzyme deactivation. Effects of mass transfer on fractional conversion and operational stability were analyzed in a fixed-bed reactor.
