High Cell Density Cultivation of Anchorage-Dependent Cells Using a Novel Macroporous Cellulosic Support

We have developed a novel cell support made of macroporous cellulose fibers (SIC) for the culture of anchorage-dependent animal cells. We examined several anchorage-dependent cell lines and found that all of them attached to the support and grew, although the efficiency of cell adsorption to the support differed with cell line. Among the cell lines tested, simian virus 40-transformed human endothelial cell line #3 was immobilized on the support and cultured in a fluidized-bed bioreactor at a high cell density. By direct introduction of air and optimization of the medium feed rate, the cells could grow to 2×10^7 cells per cm^3-SIC and be maintained over 80 d.
