Monitoring and Control of Cellulolytic Anaerobe Culture by Using Gas Evolved as an Indicator

On the culture of Ruminococcus albus in a cellulose medium, the amount of cellulose digested was proportional to that of gas evolved. Changes of substrate, cell mass and product concentrations during the culture were well simulated by the unstructured model, and some critical parameters of the model were determined from the pattern of the gas evolution. As the amount of the gas evolved can be measured with a gasmeter with a pulse generator, the culture could be monitored immediately. With the apparatus with a pulse counter and two timers, it was possible not only to automate the semi-continuous culture of a cellulolytic anaerobe, but also to maintain good control with regard to cellulose digestibility and metabolite productivity.
