Kinetics of Enzymatic Hydrolysis of Lipids in Biphasic Organic-Aqueous Systems

We proposed kinetic model for enzyme reactions in a biphasic organic-aqueous system. The rate equations were derived based on a few simple assumptions and by taking into account the interfacial area between the two phases of organic and aqueous solutions and the fraction of the organic solvent phase. Beef tallow was hydrolysed by lipase in various organic-aqueous systems to confirm the propriety of the rate equations. To analyse the rate data in more detail, the interfacial area between the organic and aqueous phases were measured by taking the microscopic photograph of the droplets of organic phase. The rate data were well represented by the rate equations derived in this paper. Kinetic parameters were found for various operating conditions, and the facilitating effects of addition of organic solvent on lipid hydrolysis are discussed.
