Monitoring and Control for Extractive Fermentation of Clostridium acetobutylicum

In the acetone-butanol fermentation using Clostridium acetobutylicum IAM 19012,it was necessary not only to keep butanol concentration below the toxic level (2 g/l), but also to control glucose concentration at less than 80 g/l and pH between 4.5 and 5.5. The amount of glucose consumed could approximately be estimated as four times the volume of gas evolved, and butanol was produced from glucose with the average yield of 0.173. It was thus possible to estimate the concentrations of glucose and butanol at any fermentation time using the volume of gas evolved as an indicator. As oleyl alcohol (cis-9-octadecen-1-ol) was an excellent extracting solvent for butanol, a fed-batch culture system for the microorganism was developed, where withdrawing and feeding operations of the solvent were done automatically based on the gas evolution.
