A pharmacognostical and chemical studies on the herbal stem of Ephedra gerardiana WALL, of Ephedraceae from Nepal were conducted in order to find out the changes of external and internal structures, and alkaloid contents, according to geographical variations. As the results, the external and internal structures showed wide variations even on special characteristics that have been believed as for its specific diagnosis, and its alkaloid contents also showed a wide range of variations. In general, the plant growing on the dried and poor soil, such as on the rock outcrop, had shorter internode and higher alkaloid content. Moreover, on the same slope the plant growing at higher elevations had shorter internode and higher alkaloid content. In addition, the node which has been anatomically defined by us contained alkaloid in the ratio of one eighth to that in the internode.
