Processing of Nux Vomica. IV. : A Comparison of Cytotoxicity of Nine Alkaloids from Processed Seeds of Strychnos nux-vomica on Tumor Cell Lines

Of the alkaloids isolated from the processed seeds of Strychnos nux-vomica, isostrychnine N-oxide and isobrucine N-oxide showed the most potent cytotoxicity to the tumor cell lines of K562, HeLa and HEP-2. The IC_<50> values of the former were 9.0 and 9.7 μM. against HeLa and K562, respectively, and that of the latter was 4.3 μm against HEP-2. These values were lower than those of the major alkaloids, strychnine and brucine. Since these compounds were formed by N-oxidation and/or ring-opening reaction of strychnos alkaloids during the processings, such as parching in a sand bath or frying in oil, both of these processing methods may be appropriate when nux vomica is to be used for possible treatment of cancer in traditional Chinese medicine.
