In the course of a survey of antiplaque agents for dental caries prevention, eighty-six different crude drugs were tested for inhibitory action in the adherence of heat-treated cells of Streptococcus mutans to glass surfaces in the presence of crude glucosyltransferase (GTase, EC 2.4.1.5) and sucrose. Eleven of the aqueous extracts, nineteen of the 50% methanolic extracts and twentytwo of the methanolic extracts showed inhibitory action at a concentration of 0.5-1.0 mg/ml. Among these, further investigation was conducted on the active methanolic extract of Artemisia capillaris spikes. Through fractionation of the extract by monitoring the ability to inhibit the cell-adherence to glass surfaces, rhamnocitrin and capillarin were isolated as inhibitory substances. This anti-plaque action was ascribed to inhibition against GTase. Free flavonols from other botanical sources also showed inhibition against GTase, but flavonol-3-glycosides did not show any appreciable inhibition.
