Metabolism of Sweroside from Swertia japonica by Human Intestinal Bacteria

Sweroside, a seco-iridoid glucoside isolated from Swertia japonica (Sen-buri in Japanese), was converted to cis-4,4a-5,6-tetrahydro-5-hydroxymethyl-6-methyl-1H,3H-pyrano(3,4-c)pyran-1-one by anaerobic incubation with human intestinal bacteria. The metabolite is considered to have been produced by the hydrolysis of sweroside by bacterial β-glucosidase, followed by a rearrangement of the aglycone and the subsequent reduction of the resulting aldehyde group to produce a hydroxymethyl group in the molecule.
