Mass Production of Lipids by Lipomyces starkeyi in Microcomputer-Aided Fed-Batch Culture

Lipomyces starkeyi was cultivated to produce a large amount of lipids by means of a microcomputer-aided fed-batch culture technique. During the cultivation, temperature, dissolved oxygen concentration and ethanol concentration in the culture broth were maintained at 30℃, 2.5±0.6 ppm and 2.5±0.3 g/l, respectively. The microbe intracellularly acuumulated a large amount of neutral lipids, when ammonium, zinc, or ferrous ion in the broth became deficient. For example, when ammonium ion in the broth was deficient at 48 h of cultivation, high cell (153 g/l) and lipid (83 g/l) concentrations were obtained at 140 h. The total of lipids produced was about twenty-fold as much as the highest previous value. Lipid yield from ethanol (g lipids/g ethanol) was 0.21 and the fatty acids of the lipids were mainly palmitic and oleic acids.
