Threonine Production from Glycine and Ethanol by a Methanol-Utilizing Bacterium

Strain KM 193,a facultative methylotroph identified as a Pseudomonas sp., produced threonine from glycine and ethanol in a reaction mixture using resting cells grown on methanol. Methanol-grown cells of this strain also produced serine from glycine and methanol. To obtain cells of strain KM 193 with high threonine-producing activity, we investigated optimal cultural conditions. The reaction products from glycine and ethanol were L-threonine and allothreonine. The optimal reaction conditions for threonine production by resting cells of this strain were found. A glycine-resistant mutant G3 derived from strain KM 193 produced more L-threonine and L-serine : 0.31 mg/ml of L-threonine, 1.5-fold that from the parent, and 2.5 mg/ml of L-serine, 2.3-fold that from the parent. The mutant had serine transhydroxymethylase activity nearly 1.7-fold that of the parent. Air-dried cells of strain KM 193 also had produced threonine, but not L-serine. Cell-free extracts synthesized L-threonine and L-serine from either the correponding alcohol or aldehyde.
