Formation of Valine and Leucine by Analog-resistant Mutants of an Obligate Methylotroph, methylomonas aminofaciens

• Norleucine-, norvaline-, and valine hydroxamate-resistant mutants were derived from Methylomonas aminofacients to get improved ability of valine and leucine formation. Using a norvaline-resistant mutant, M325,various culture conditions for the amino acid formation were examined. When methanol was fed proportionally with the increase of growth, strain M325 formed larger amounts of the amino acids than by conventional batch culture. The potassium phosphate concentration and the addition of some organic acid affected the amino acid formation. Especially, pyruvate and thiamine promoted it. On cultivation in medium containing pyruvate, thiamine, and 2% methanol, a valine hydroxamate-resistant mutant, X329,formed 2.2 mg/ml of L-valine and 0.8 mg/ml of L-leucine.
