L-Serine Production by Resting Cells of a Methanol-Utilizing Bacterium

Among about 400 methanol-utilizing microorganisms, a bacterial strain, KM 146,was selected as a serine producer from glycine and methanol with a resting cell system. A large-scale preparation method for resting cells of strain KM 146 was established by investigating the culture conditions including medium composition, methanol and mineral salts feeding and pH control. Under suitable culture conditions, 74 mg (as dry weight)/ml of cells having high serine-producing activity were obtained in a 10-day culture broth. With the resting cells of strain KM 146,the reaction conditions for L-serine production were optimized. The optimum pH and temperature were 8.2 and 30℃, respectively. The reaction needed appropraite aeration. Methanol feeding during the reaction enhanced the L-serine production. Under the optimum conditions, 24 mg/ml of L-serine was produced from 100 mg/ml of glycine. The L-serine produced in the reaction mixture was isolated and identified.
