Amino Acid Formation by Methanol-utilizing Bacteria

About 400 methanol-utilizing microorganisms isolated from soil were examined for their amino acid accumulation. When strain No. 15a was grown in medium initially containing 1% methanol, with further addition of methanol and urea to 1% and 0.025%, respectively, after 2 and 3 days cultivation, about 0.25 mg per ml of alanine were formed after 4-day culture. Using another strain, No. 77a, the cultural conditions for amino acid formation were investigated, including initial methanol concentration, feeding of methanol and urea, nitrogen source and initial pH of medium. Under the conditions tested, the organism formed about 0.6 mg per ml of valine and 0.4 mg per ml of leucine after 8-day culture. The valine and leucine formed were isolated and identified. From taxonomical studies, strain No. 77a, which utilizes only methanol as the sole carbon and energy source among the tested organic compounds including various C_1 compounds, was named Methylomonas aminofaciens nov. sp.
