Fermentation and Metabolism of Nucleic Acid-related Compounds in Yeasts

Enzyme preparations of Baker's yeast and Saccharomyces sake phosphorylated AMP to ADP and ATP accompanying with glucose degradation. ADP and ATP were isolated from the reaction mixture. It was observed that phosphate concentration affected on the metabolism of AMP. In low phosphate concentration, AMP was degraded by adenylic deaminase, inosine or adenosine nuc1eotidase and hypoxanthine nucleosidase to form IMP, inosine, adenosine and hypoxanthine. In high phosphate concentration, AMP was degraded to adenosine and adenine. With adequate amount of phosphate, AMP was converted to ADP and ATP. Accumulation of IMP was observed when large amounts of AMP (30〜45μmoles/ml) were added in the reaction mixture. The mechanism of this phenomenon was discussed. Degradation of AMP was studied. Properties of adenylic deaminase were investigated using cell-free extracts and partially purified enzyme preparation of Baker's yeast. It was observed that this enzyme was inhibited competitively by inorganic phosphate and that this inhibition was released by increment of substrate concentration. This enzyme was specific to AMP. Enzymic degradation of IMP was discussed. It was found that degradation of IMP was inhibited by inorganic phosphate and coexistence of AMP. It was observed that AMP had an effect in releasing the glycolysis inhibition caused by high concentration of salt and had some promoting function on glycolysis.
