Unique Production of a Disaccharide, Galβ1→3GalNAc, by Endo-α-N-Acetylgalactosaminidase of Alcaligenes sp.

An enzymic method for producing a disaccharide was developed using the endo-α-N-acetylgalactosaminidase from Alcaligenes sp. From 0.5 g asialofetuin, 2.66 mg of pure disaccharide was obtained by procedures including enzymic hydrolysis, alcohol fractionation, gel filtration on Sephadex G-10 and normal-phase high-performance liquid chromatography on Cosmosil 5 NH_2. The structure of the disaccharide was determined as Galβ1→3GalNAc by thin -layer chromatography, gas-liquid chromatography and proton nuclear magnetic resonance spectroscopy. The molecular extinction coefficients of the disaccharide at 210 nm, 200 nm and 190 nm were found to be 1.17×10^3 M^<-1>, 4.36×10^3 M^<-1> and 8.00×10^3 M^<-1>, respectivery, and he molecular extinction coeficient of its derivative at 585 nm was determined as 15.6×10^3 M^<-1> by using the method of Reissig et al. (J. Biol. Chem., 217,959-1955). This method of producing the disaccharide is considered to be applicable for practical use.
