Properties of Brevibacterium flavum Glutamine Synthetase in an "in vivo-like" System

• An "in vivo-like" reaction mixture with an ATP-regenerating system was estabilshed to reexamine the properties of glutamine synthetase from Brevibacterium flavum. In the mixture, glutamine synthetase activity with an optimal concentration or Mn^<2+> was about 3 times higher than that with Mg^<2+> and the apparent K_m for Mn^<2+>-ATP was much lower than that for Mg^<2+>-ATP. The kinetic response to ammonia showed biphasic curves when Mg^<2+> was used as a metal ion, but not when Mn^<2+> was used. Both the Mg^<2+>-dependent and the Mn^<2+>-dependent reactions were not so susceptible to some amino acids as in the standard assay mixture containing higher concentrations of the substrates. As for the effects of AMP, adenosine and adenine, the difference between Mg^<2+>-dependent and Mn^<2+>-dependent reactions was more marked than in the standard assay mixture.
