Isolation and Purification of Choline Kinase from Hansenula jadinii

Choline kinase activity from dried cells of Hansenula jadinii IFO 0987 was separated into three distinct fractions, choline kinases A, B and C, by DEAE-cellulose column chromatography with a concentration gradient of ammonium sulfate in 0.1 M potassium phosphate buffer. Two fractions, choline kinases B and C, were further purified approximately 100- and 390-fold by hydroxylapatite column and Sephadex G-150 gel chromatography. Crystalline preparation of choline kinases B and C were obtained by ammonium sulfate saturation. They had different isoelectric points of pH 5.0±0.1 and 4.1±0.05.
