NADH production from NAD^+ using a Formate Dehygdogenase System with Cells of a Methanol-Utilizing Bacterium

A facultative methanol-utilizing bacterium, Arthrobacter sp. KM 62,produced NADH from NAD^+ in a formate dehydrogenase reaction system in which freeze-thawed, air-dreid or acetone-dried cells were used. Intact cells of the bacterium also showed high NADH production when some surfactants were added to the reaction mixture.Using air--dried cells of strain KM 62,optimum conditions for NADH production were examined, including formate concentration, NAD^+ concentration, pH, buffer type and reaction temperature. Under suitable conditions the conversion ratio and the amount of NADH accumulated reached 90% and about 30 mg/ml of the reaction mixture, respectively. The NADH produced in such a reaction was purified in powdered form with 83% purity as judged from the ratio of absorbance at 340 nm to that at 260 nm. The purity was confirmed to be 90-91% from the coenzyme activity with NAD-dependent alcohol dehydrogenase.
