Enzymatic Preparation of Crystalline Glucose from Waste Cellulosic Materails

This paper is concerned with enzymatic preparation of crystalline glucose from saccharification-residue which was the solid materials after enzymatic removal of hemicellulose from waste cellulosic materials such as corncobs and rice straw.For a typical experimental preparation of crystalline glucose, 200 g (dry matter) of corncobs was pretreated with 2% NaOH solution at 30℃ for 6h, and the xylan in the alkali-treated material was hydrolytically removed with a xylanase. The saccharification-residue (80.8 g of dry matter) thus obtained was hydrolyzed with 16 g of Cellulase AP produced by Aspergillus niger, at 50℃ and pH4-4.2 for 4 days. The resulting sugar solutin, containing 53.0 g of total reducing sugar, was decolorized, desalted and concentrated. Crystalline glucose (41.4 g) was obtained.The existence of oligosaccharides such as gentiobiose and xylobiose was confirmed in the molasses.
