Inducers for Xylanase Production by Crytococcus flavus

Cryptococcus flavus produced a xylanase inductively when grown in a medium contained xylose, xylooligosaccharide, or xylan. To obtain a more effective inducer, a variety of derivatives of xylose, xylobiose, and xylan were tested for their inducing ability. It was found that β-methylxyloside was a non-metabolizable inducer and possessed a higher inducing ability than other materials. By the addition of the β-methylxyloside (0.3 mg/ml) to a glucose medium, the xylanase activity in the culture medium increased about 15-20 times that obtained in xylose or xylan media. The uptake of β-xylosides by cells grown on glucose medium was selective depending on differences of the aglycon structure of these β-xylosides. It was found that almost all of the β-methylxyloside added to the inducing culture medium was accumulated in the cell against a concentration gradient. However isopropyl-, butyl-, benzyl-, and thiophenyl-β-xyloside were not taken up by the cells, so that xylanase induction did not occur.
