To search for possible anti-tumor-promoters and cancer chemopreventive agents, we carried out a primary screening of several Panax plants using an in vitro synergistic assay system. The MeOH extract of the root of Panax notoginseng exhibited a significant inhibitory effect on Epstein-Barr virus (EBV) activation induced by a tumor promoter, 12-O-tetradecanoylphorbol-13-acetate (TPA). The major saponin of the roots of P. notoginseng, ginsenoside Rg_1, also exhibited a potent inhibitory effect on EBV activation. Furthermore, an extract of this plant exhibited the strong anti-tumor-promoting effects on the two-stage mouse skin (7,12-dimethyl-benz [a] anthracene as an initiator and TPA as a promoter), pulmonary (4-nitroquinoline-N-oxide as an initiator and glycerol as a promoter) and hepatic (N-nitrosodiethylamine as an initiator and phenobarbital as a promoter) carcinogenesis tests in vivo.
