The culture condirions for accumulation of vitamin B_<12> by Pseudomonas AM-1 and Microcyclus eburneus in methanol medium and the form of the vitamin produced were investigated. Addition of organic nutridnts such as meat extract, casamino acid and polypeptone promoted vitamin B_<12> formation. Optimal concentrations of methanol and cobalt chloride were 2%(v/v) and 2 mg/l, respectively. The organisms utilized methanol, ethanol, glucose, glycerol and 1,2-propanediol. Ps. AM-1 produced vitamin B_<12> only on methanol or propanediol, while Mic. eburneus produced much vitamin B_<12> on all these substrates.
