RAPD Analysis of Ephedra Plants

Genomic DNAs from fresh stem of Ephedra plants and crude drug "Mahuang" were studied by random amplified polymorphic DNA (RAPD ) analysis. The dendrogram retrieved from the RAPD profiles revealed that 9 samples from fresh stem were grouped into 3 clusters, Cluster A, Cluster B and Cluster C. The sample from the crude drug connected to the Cluster A although its dissimilarity value was larger. This clustering agreed with the results of chloroplast chlB gene sequences. It is reasonable to conclude that we are able to classify the Ephedra plants used in this study by RAPD analysis. Some of the label names used in each medicinal plant garden did not agree with the classification based on molecular traits. This result suggests that the label names of Ephedra plants in the medicinal plant gardens of Japan, including the plants used in this study, are in a state of confusion.
