Production of Anethole in Callus of Illicium verum HOOKER filius

Callus tissue was induced from a leaf of Illicium verum Hooker filius (Illiciaceae) in a half strength salt formulation of Woody Plant (1/2 WP) medium solidified with 0.25% of gelrite, containing 2% of sucrose and combinations of 1-naphthaleneacetic acid-6-benzylaminopurine or kinetin-2,4-dichlorophenoxyacetic acid. The optimum medium for the callus growth was found to be 1/2 WP medium containing 5 mg/l of kinetin and 1 mg/l of 2,4-dichlorophenoxyacetic acid. A diethyl ether extract of the callus contained anethole and foeniculin as the major components. The anethole and foeniculin contents of callus were the highest when the callus was incubated in 1/2 WP medium supplemented with 1 mg/l of 1-naphthaleneacetic acid and 5mg/l of 6-benzylaminopurine, and the foeniculin content was always higher than the anethole content. The present paper reports the callus induction from an I. verum leaf and the anethole production by the callus tissue.
