Induction and Rapid Propagation of Shoot Primordia of Mentha arvensis L. var. piperascens by Shoot Tip Culture

Shoot primordia were induced from the apical domes of shoot tips of Mentha arvensis L. var. piperascens (Labiatae) on Murashige and Skoog (MS) liquid medium supplemented either with 6-benzyl-aminopurine (BAP) 10^<-6>, 10^<-5>, 2×1O^<-5> M or with α-naphthaleneacetic acid (NAA) 10^<-7> M plus BAP 10^<-6> M. The cultures were shaken at 2 cycles per minute on a rotary culture equipment, under light. The shoot primordia were capable of good vegetative propagation and easily developed into normal plantlets within 1 month, either on hormone-free MS or Gamborg-B5 (G-B5) static media solidified with 0.2% gelrite. Further shoot primordia of M. arvensis var. piperascens were induced on G-B5 liquid medium supplemented with BAP at a concentration of 10^<-5> M or 2×10^<-5> M. For this experiment, the shoot tips were taken from in vitro regenerated plantlets on MS solid media through the shoot primordia, which were initially developed on MS liquid media.
