Formation of Stigma-like Structures and Pigments in Cultured Tissues of Crocus sativus

Excised young stigma and style tissues of Crocus sativus cultured in vitro formed style-like structures with terminal stigma-like tips. The tip portion of the stigmas did not have such developmental abilities. For this proliferation, the presence of 6-benzylaminopurine (BAP) (10^<-5>-5×10^<-5> M) and α-naphthaleneacetic acid (NAA) (10^<-5>-3×10^<-5> M) in the medium was necessary. The optimal concentrations of NAA and BAP for the proliferation were 5×10^<-5> M and 3×10^<-5> M, respectively. The organogenesis was induced in the dark at 25 ± 2℃. Chemical analyses showed the presence of both crocin (yellow pigment) and picrocrocin (bitter glycoside) in these tissues. This indicated that the proliferating stigma-like and style-like structures in vitro synthesized stigma-specific components as are found in intact stigmas.
