Propagation of Bupleurum falcatum by Shoot Tip Culture : Induction of Shoot Primordia

Shoot tips of Bupleurum falcatum (2n = 20) L. (Umbelliferae) were cultured in 27×200 mm culture vials with slow shaking at 2 rpm on a gyrated drum placed obliquely. Shoot primordia were induced in Murashige-Skoog (MS) liquid medium containing 6-benzylaminopurine (BAP) and α-naphthaleneacetic acid (NAA) under light. The propagation of the shoot primordia of B. falcatum was rapid and they were highly stable in the chromosome number and karyotype. By static culture on the hormone-free Gamborg B5 (B5) medium solidified with 0.2% gerlite, the shoot primordia readily developed into plantlets with shoots and roots within 5 weeks. The plantlets were transplanted in sterilized soil running fields to grow to normal adult plants.
