Medium-term Conservation of Scopolia japonica Shoot Cultures and Evaluation of the Regenerated Plants

In this paper, we present a protocol for ensuring the cold storage of shoot cultures of Scopolia japonica Maxim., a medicinal plant of the family Solanaceae containing hyoscyamine and scopolamine. and the results of subsequent biological and biochemical evaluations of plants raised from cold-stored in vitro shoots. All and 70 % of the shoot cultures withstood storage at 2℃ for one and two years, respectively. No shoots, however, could survive cold storage for three years. Shoots cold-stored for two years were multiplied on freshly prepared Linsmaier and Skoog medium supplemented with 1 μM indole-3-acetic acid and 10 μM 6-benzyladenine, and rooted in the same basal medium containing 1 μM indole-3-acetic acid under normal culture conditions. A high percentage (97 %) of rooted plants could be acclimatized and they were cultivated outdoors for three or four years. No essential changes were found in their morphological characteristics or in the composition and contents of tropane alkaloids in the rhizome when compared with the control set. These findings demonstrate that the cold storage of in vitro shoot cultures can be used as a reliable method for medium-term conservation of some medicinal plants.
