Effects of Gelling Agents on the Growth and Naphthoquinone Accumulation of Lithospermum erythrorhizon Callus Cultures

Five gelling agents were tested for their effect on the growth of and the metabolite accumulation in callus cultures. Of five culture lines tested, only in the callus culture of Lithospermum erythrorhizon the accumulation of naphthoquinone pigments was significantly enhanced by two of the gelling agents Gelrite (Kelco) and Purified Agar (Difco). In this case, the enhancement appeared to be caused by the difference in the chemical composition of the gelling agent and not by the difference in the medium hardness. Other callus cultures and metabolites tested were: Bupleurum falcatum-anthocyanin; Mallotus japonicus-chlorophyll a and b; Nicotiana tabacum-nicotine; and Phytolacca americana-betalains, in which little change was observed in the growth or metabolite accumulation by the use of different gelling agents.
