Morphological and Chemical Evaluation of the Plants Regenerated from Cold-Preserved In Vitro Shoots of Datura metel L.

Shoot cultures of Datura metel var. rubra and D, metel var. fastuosa were established on Linsmaier and Skoog medium with 1μM 6-benzyladenine and successfully preserved at 15℃ for 62 weeks. The shoots were recultured on the same medium under normal culture conditions and rooted normally on Linsmaier and Skoog medium containing 1μM indole-3-butyric acid. The regenerated plants grew well in pots. Morphological features and tropane alkaloid composition of the regenerated plants were comparable to those of the control. These findings show that cold storage of in vitro D. metel shoots at 15℃ can be used as a germplasm conservation system for medium-term duration without deterioration of the ability to accumulate tropane alkaloids.
