Gastrin-stimulated Gastric Acid Secretion while H_2-receptors are Blocked by an H_2-receptor Antagonist

Gastrin-stimulated gastric acid secretion was studied not only by gastric analysis but also by the endoscopic Congo-red test using six mongrel dogs as materials while H_2-receptors were blocked by cimetidine, an H_2-receptor antagonist. Remarkable inhibition of gastrin-stimulated acid secretion by cimetidine were observed in the gastric analysis. These effects were also confirmed endoscopically by the Congo-red test. The discoloration into bluish black was remarkably suppressed by cimetidine with respect to speed, the depth of color and the size of the area. Even 20min. after the start of the Congo-red test, the discoloration was uneven without the appearance of border-lines between the bluish black zone and red zone in cases of cimetidine administration, while uniform discoloration was observed with a clear boder-line in cases without administration of cimetidine. These data suggest that the mechanisms increasing the availability of histamine play a major role in acid secretion in response to gastrin or that the receptor-site of gastrin is strongly blocked when the receptor-site of histamine is blocked by H_2-receptor antagonist. The phenomena observed in the Congored test suggest that the density or activity of parietal cells are unequal in the stomach.
