Surface structures of human colorectal mucosa and polyps were observed using dissecting microscopy and scanning electron microscopy (SEM). The specimens were obtained by endoscopic biopsy and polypectomy from the large intestine. Careful manipulation was an important aid in such procedures as fixation, dehydration, drying and surface coating for observations of the intestinal mucosal fine structure. The pictures in dissecting microscopy were magnified within the low power range, and scanning electron microscopy was able to magnify from the low power to high power ranges. In this paper, it is reported that the surface appearances of normal colorectal mucosa revealed a honey-comb pattern and the openings of the crypt of the colorectal polyp showed various sizes and shapes. By dissecting microscopy and SEM, the surface structure of the intestinal mucosa was also examined satisfactorily even in biopsy and polypectomy specimens. Our procedures are also valuable in the study of the fine surface structure of the digestive tract.
