Phosphatidylglycerol Synthesis by Phospholipase D in a Microporous Membrane Bioreactor : Studies on Enzymatic Conversion of Phospholipids (I)

Transphosphatidylation of lecithin to phosphatidylglycerol by phospholipase D was done using a specially designed microporous hydrophobic membrane bioreactor. The enzyme was purified six-fold from cabboge leaves. Several ethers were effective for stabilizing phospholipase D in the bioreactor, but the enzyme solution without solvent was quite unstable. Its half life was calculated as 7.5 h in the enzyme assay. The higher concentrations of glycerol were effective for phospholipase D stability, but such enzyme solutions gave lower activities. The optical concentration of glycerol for the synthesis of phosphatidylglycerol was between 10% and 20% in this bioreactor. This means that at lower concentrations of glycerol, much more phosphatidylglycerol was synthesized than at higher glycerol concentrations, so the synthesis of phosphatidhlglycerol in transphosphatidylation was not proportional to hlycerol concentration. Moreover, the phosphatidylglycerol formation was higher at a low concentration of lecithin. The advantages of phosphatidylglycerol synthesis by the bioreactor are that (1) the interfacial area is constant, (2) this system does not need a surfactant nor stirring during the reaction, (3) the products obviate the need of separation from enzyme solution, (4) minimization of autoxidatioin of substrate and enzyme, (5) the recycle system economizes the enzyme, and (6) continuous operation is possible if it is desired for an industrial system.
