755 Monitoring of lipase-catalyzed transesterification between eicosapentaenoic acid ethyl ester and tricaprylin by silver ion high performance liquid chromatography and high temperature gas chromatography

A high speed, high resolution approach to silver ion HPLC and high temperature gas chromatography with an on column injection methods have been established for the separation of triacylglycerols(TAGs) which contain eicosapentaenoic acid (EPA) molecules at different positions of TAGs. Using a solvent system based on n-hexane, 2-propanol, and acetonitrile, the separation of TAG containing EPA depending on the number of EPA molecules and their isomeric configuration could be obtained. This technique was able to be applied to monitor the incorporation of polyunsaturated fatty acid (PUFA) molecule to the specific position of TAG. For the monitoring of the synthesis of structured TAG containing EPA, transesterification of tricaprylin with EPA ethyl ester (EPAEE) in a solvent free system was performed. The transesterification reaction by various lipases at a different molar ratio of substrates, at several initial water activity (a_w) of lipases, and at some reaction temperature conditions were investigated.
