Stimulatory Effect of Ammonium on Streptomycin Formation by Streptomyces griseus Growing on a Glucose Minimal Medium

Streptomyces griseus was cultivated in a pH-stat batch culture on a glucose-ammonium minimal medium with varying initial ammonium concentration. A stimulatory effect of ammonium on streptomycin formation was observed in the idiophase, i.e., the amount of streptomycin formed in the idiophase were 0(initial ammonium sulfate, 2g/l), 62(4 g/l) and 104 mg/l(5 g/l), although the amount of streptomycin at the end of the trophophase was about 37 mg/l in all the cultures. The effect of ammonium on streptomycin formation coincided with the overproduction of glutamic acid during the trophophase and it was consumed prior to streptomycin formation in the idiophase. In addition, when glutamic acid, glutamine and glucosamine were respectively supplemented to the culture at the end of the trophophase, streptomycin formation in the idiophase could be stimulated. Based on these results, the role of glutamic acid and glutamine in streptomycin formation, serving as an amino-group donor in the biosynthetic pathways of streptomycin, was discussed.
