Enzymatic Reduction of cystine into Cysteine by Cell-Free Extract of Clostridium thermoaceticum

Cell-free extract from an acetate-tolerant thermophilic homoacetogen, Clostridium thermoaceticum catalyzed the reduction of L-cystine into L-cysteine in the presence of molecular hydrogen and methyl viologen (MV). The optimum reaction conditions for the reduction of cystine into cysteine were pH 9.5 and a temperature of 60℃. The fact that cystein was not detected in the absence of cell-free extract, MV or molecular hydrogen proved that the reduction of cystine into cysteine was coupled with the reoxidation of a reduced form of MV, which was established through the hydrogenase-catalyzed reduction of the oxidized form of MV with hydrogen. Seventy five mM of cystine was completely converted to 150 mM of cysteine using 15 units of hydrogenase in cell-free extract, 0.5 mM of MV and 100% hydrogen in the gas phase for 6 h at pH 9.5 and at 60℃.
