Mutual Conversion between H_2 Plus CO_2 and Formate by a Formate-Utilizing Methanogen

Formate dehydrogenase, hydrogenase and F_<420>-NADP^+ oxidoreductase could be measured in a formate-utilizing methanogen, methanogen strain HU, anaerobically growth on a formate minimal medium.Formate could be manly converted to methane with a slight evolution of hydrogen, in pH ranges between 5.5 and 8.5 obeying the theoretical conversion ratio of formate to methane. Slight evolution of hydrogen was observed at the early stage of fermentation especially in acidic(pH5.5)or alkaline(pH8.5)conditions.With chloroform, an inhibitor of methly coenzyme M reductase and methanogenesis, in the culture medium, either hydrogen evolution from formate as substrate or formate formation from hydrogen plus CO_2 as substrate could occur, suggesting that the formate hydrogenlyase system might operate reversibly in this bacterium.
