Isolation of a Formate-Utilizing Methanogenic Bacterium by a Chemostat Culture Technique

A formate-utilizing methanogen has been isolated from an anaerobic digester by means of a chemostat culture using formate at the sole carbon source. This technique was efficient for the purpose of isolating the methanogens because of their unique ability to grow on a formate minimal medium under strictly anaerobic condition. Eventual contaminants and the slowly growing methanogens could be washed out from the fermentor at high dilution rates and consequently, the colony isolation of a high efficient methanogen on anaerobic agar plates could be accomplished using a well acclimatized methanogenic culture. The methanogen strain HU we isolated was a strictly anaerobic, Gram-negative, motile rod. The DNA base composition was 61.3 mol% guanine puls cytosine. The optimum pH and temperature estimated from the formate consumption and methane production were 7.2 and 42℃, respectively. Formate or hydrogen and carbon dioxide could serve at the sole carbon source for growth but methanol and acetate could not be utilized by this organism.
