Requirements of PO_4^<3->, NO_3^-, SO_4^<2->, K^+, and Ca^<2+> for the growth of Tabacco Cells in Suspension Culture : Biomass Production of Tobacco Cells(III)

During the growth of tabacco cells (Nicotiana tabacum L. c.v. BY-2) in suspension culture in sucrose medium under dark-aerobic conditions, phosphate ion (PO_4^<3->) in the basal medium was almost completely consumed at the initial stage of batch culture, although cells continued to grow well after PO_4^<3-> had disappeared from the medium. The increase of KH_2PO_4 in the medium from 340 to 510 mg/l resulted in increase of specific growth rate from 0.80 to 0.96 per day. The growth yields for sucrose and major inorganic constituents measured during exponential growth phase were Y_<X/S>=0.45 (g cell/g sucrose), Y_<X/NO_3^->=6.9 (g cell/g NO_3-), Y_<X/SO_4^<2->>=107(g cell/gSO_4^<2->), Y_<X/K^+>=40.3(g cell/g K^+) and Y_<X/Ca^<2+>>=356(g cell/g Ca^<2+>). A medium optimization was attempted from the information obtained.
