Physical properties such as viscosity, density and fluid behavior of the culture broth, and cell shape of tobacco cells induced from Nicotiana tabacum L. c.v. Bright Yellow-2 (BY-2) were studied.Broth viscosity measured by rotation viscometer at 60 rpm increased remarkably after 49.5 hr of cultivation and reached the maximum at 74 hr, which was 27.5 times higher than the initial value. The viscosity of culture filtrate, however, changed only from 0.9 to 2.2 cp during the cultivation. Though the culture filtrate acted as a New-tonian fluid, the culture broth exhibited pseudo-plastic behavior. The flow behavior index, n, varied slightly from 0.69 to 0.74 during the cultivation.Changes in the size of tobacco cell aggregates, which seemed to have some influence on broth viscosity, were measured during the cultivation. In the injtial stage of batch culture, cells were predominantly dispersed; as the culture progressed, aggregations formed, for example, of 350〜400 μm (maximum class interval) after 3 days; and towards the end of the cultivation, i.e. after 5 days, the aggregates started to break up.
