Biomass Production of Nitrogen-reduced Tobacco Cells in Two-stage Continuous Culture : Biomass Producito of Tobacco Cells(V)

Continuous culture of tobacco cells of Nicotina tabacum L. was carried out in 60-l fermentors for biomass production for use of them as tobacco raw material. For the production of nitrogen content-reduced tobacco cells in continuous culture, both single-stage and two-stage cultuers were carried out. In the two-stage fermentaiton system, the first fermentor was fed the basal medium and the second fermentor was fed the medium with the nitrogen content decreased to 1/5 of that in the basal medium together with culture broth from the 1st fermentor. Biomass concentrations and productivity of cells in the two-stage fermentaiotn were X_1=15.1 g/l and X_2=13.6 g/l, and D_1X_1=7.9 g/l/day and D_2X_2=13.2 g/l/day (subscripts represent the stages of continuous culture). Intracellular total nitrogen content in the 1st and the 2nd fermentors was 4.6% and 3.4%, and protein nitrogen was 3.8% and 2.9%, respectively, at high dilution rates of D_1=0.52 day^<-1> and D_2=0.97 day^<-1>. Productivity of nitrogen-reduced cells (per one fermentor) in the two-stage continuous culture was increased by 18% compared with that in the single-stage continuous culture at D=0.42 day^<-1>. But obvious changes in the appearance of the culture such as color and aggregate were observed during continuous culture.Further two-stage continous culture was carried out changing the concentration of NH_4NO_3 in the feed medium of the 1st fermentor to 1.5 times of that in the basal medium. Using this medium improved the color of the cells and culture broth in the 1st fermentor, but was less effective for obtaining dispersed cells. Biomass yields obtained in the two-stage cultuer at D_1=0.41 day^<-1> and D_2=0.78 day^<-1> were X_1=16.1 g/l and X_2=16.1 g/l, and D_1X_1=6.6 g/l/day and D_2X_2=12.6 g/l/day.These results show that applicaiton of the two-stage fermentaiton system would allow high productivity of nitrogen-reduced tobaco cells for use as raw materials of cigarettes.
