Purification and Properties of Endo-1,4-β-Xylanase from Humicola lanuginosa

An endo-xylanase (1,4-β-D-xylan xylanohydrolase, EC 3.2.1.8) was extracted from a wheat bran culture of Humicola lanuginosa. The enzyme was purified 54-fold with 68% yield by gel filtration and ion exchange chromatography. The purified enzyme had a molecular weight of approximatery 21,000 with an isoelectric point of 4.1. The optimum pH was 6.0 and the temperature, 65℃. In xylan hydrolysis by the enzyme, xylooligosaccharides were observed at the beginning, and after prolonged incubation, xylotriose and xylobiose were predominant, with a small amount of xylose. The results suggested that this enzyme was an endo-type xylanase. However, when xylobiose was used as a substrate, a trace of xylose was observed. The apparent Michaelis constant was 7.3 mg/ml and xylobiose was shown to be a comperirive inhibitor to xylanase with a K_i of 1.4 mg/ml.
