Growth on Propionate-Acetate Media of Rhodopseudomonas sphaeroides S and Its Vitamin B_<12> Biosynthesis

Effects of Co<2+> and/or HCO_3^- on propionate utilization and vitamin B_<12> biosynthesis of Rhodopseudomonas sphaeroides S were studied under dark-aerobic conditions in propionate-basal media. Remarkable growth and intracellular accumulation of vitamin B_<12> were observed by adding cobalt ion and yeast extract to the propionate-basal media, the growth was evidently depressed without cobalt ion in the media. The addition of vitaimn B_<12> with yeast extract to the propionate-basal medium also stimulated the growth to the same extent as CO^<2+> addition. The addition of NaHCO_3 up to 1 g/l in propionate-basal medium with yeast extract stimulated the growth in direct proportion with the amount added, and the addition of NaHCO_3 with either Co^<2+> or vitamin B_<12> increased the growth about 2-fold compared with the sole addition of NaHCO_3. These facts suggest that propionate might be metabolized via the methylmalonyl-CoA pathway by this organism.When mixtures of acetate and propionate at various ratios were used in the presence of cobalt ion and yeast extract, both acids were simultaneously utilized with the accumulation of vitamin B_<12> in the cells. Intracellular accumulation of vitamin B_<12> reached a maximum, 67 μg vitamin B_<12>/g dry cell, with 1.8 g of C_2H_5COONa and 0.2 g of CH_3-COONa・3H_2O in 1 l of basal medium supplemented with yeast extract and Co^<2+>. Vitamin B_<12> was present in the cells in the form of cobalamin coenzyme.
