Growth Stoichiometries and the Efficiency of Oxidative Phosphorylation in Pseudomonas AM-1 Growing on Methanol, Formaldehyde and Formate

An attempt was made to estimate the P/O ratios for the prosthetic groups (XH_2,YH_2 and NADH_2) in Pseudomonas AM-1 aerobically growing on methanol, formaldehyde and formate as the sole carbon source. The method was as follows. Firstly, growth stoichiometries for C_1-compounds were derived from chemostat cultures with methanol or formaldehyde or formate as the limiting carbon source. Secondly, growth stoichiometries for C_1-compounds were theoretically constructed on the basis of the homoisocitrate-glyoxylate cycle coupled with the serine pathways. For the construction of the growth stoichiometries it was assumed that the synthesis of cell material from 3-phospho-glycerate (PGA) requires 0.095 mol ATP/g cell (Y<ATP>=10.5) as energy and 0.02 mol NADH_2/g cell to reduce PGA to the cell material. Thirdly, by comparing the experimentally derived stoichiometries with the theoretical ones, estimations of P/O ratio were made under the assumption that 0.33 and 0.50 mol ATP are necessary to transport one proton from formate. Consequently the P/O ratios for NADH_2,XH_2 (quinone derivative) and YH_2 were estimated to be in the ranges of 1.2 to 1.7,0.8 to 1.1 and 0.1 to 0.6,respectively, provided that Y<ATP> is from 8 to 12.
