A Fed-Batch Culture of Methanol-utilizing Bacteria with pH Stat : Studies on Methanol Metabolism(VI)

A practical fed-batch culture, in which a methanol-ammonia mixture was fed in respones to a direct signal of pH change, was carried out for the production of biomass and vitamin B_<12> using methanol-assimilating bacteria, Pseudomonas AM-1 and Klebsiella sp. No. 101.Methanol concentration in the culture broth was kept almost unchanged at an appropriate concentration for growth, 0.5% (v/v), during the exponential growth of both bacteria. Exponential growth continued for 24 hr yielding 15 g cell/l and 0.6 mg B_<12>/l with Pseudomonas AM-1,and subsequently the growth rate gradually decreased, mainly because of oxygen deficiency, to give final biomass and vitamin B_<12> levels of 36 g cell/l and 3.2 mg B_<12>/l after 85 hr cultivation.
