A possible mechanism of the pressor effect of cadmium was investigated by using the isolateed thoracic aorta from rat. Cadmium produced the contraction in the low concentration and the relaxation in the high concentration. The contraction by cadmium was not inhibited by the high concentration of phentolamine, the sympathetic α-blocking agent. The contraction was reduced in accordance with the decrease of Ca in the medium and disappeared in the Ca-free medium, rather inducing a small relaxation. When the low concentration of cadmium was applied in a short time, the contraction was remarkably reduced and finally disappeared. While, the contraction by noradrenaline was not affected by the cadmium tachyphylaxis. Cadmium at the low concentration potentiated the contractions by K, Ba and noradrenaline, while that in the high concentration inhibited them. These results suggest that the contraction by cadmium is dependent on the external Ca and that the low concentration of cadmium accelerates the Ca release from the Ca store, the high concentration inhibiting the Ca release and/or the function of contractile proteins.
