The application of K^+, noradrenaline (NA), acetylcholine (ACh), or Ba^<2+> to the vas deferens strips from guinea pig, and the addition of Ca^<2+> to the strips exposed to high-K^+ depolarizing Ca^<2+>-free media produced phasic or tonic contractions. Treatment with cadmium in low concentrations potentiated the contractile responses to these agonists. The potency for the augmented effects was in the order of K^+>Ba^<2+>>Ca^<2+>>ACh = NA. While, high concentrations of cadmium suppressed the agonists-induced contractions in a dose-dependent manner. The potency for the inhibitory effects was in the order of ACh = NA>Ca^<2+>>Ba^<2+>>K^+. A stronger inhibition was also shown against the tonic contraction than the phasic contraction produced by K^+. The findings suggest that the potentiating mechanism for cadmium in low concentrations contributes to the increase of Ca release from the intracellular storage sites and Ca influx across the cell membrane, whereas that the inhibition by cadmium in high concentrations relates to the interference of those caused by the agonists.
