Contractile Mechanism of ATP in Isolated Vas Deferens of Guinea Pigs

The contractile mechanisms of ATP and the related compounds were investigated in the isolated vas deferens of guinea pig. ATP and ADP produced contractions, while AMP and adenosine had no effect. The effect of ATP was about twice as potent as that of ADP. The contraction by ATP was slightly inhibited by atropine and phentolamine, but it was potentiated by eserine. The shape of the contraction was phasic and the change of membrane potential was also transient. From these results, it is suggested that the contraction by ATP is due to the indirect stimulation to ACh and adrenaline receptors by ACh and catecholamine released from the endings of cholinergic and adrenergic fibers in small part and the direct stimulation to cell membranes in large part, and the shape of the contraction is in close connection with the change of membrane potential.
